چکیده

Recent studies provide evidence that phospholipase A2 (PLA2) may play a role in the development of experimental parkinsonism. In this investigation an attempt was made to determine a possible protective effect of quinacrine (QNC), a PLA2 inhibitor on MPTP as well as 6-hydroxydopamine (6-OHDA)-induced neurotoxicity in rodents. For MPTP studies, adult male mice (C57 BL) were treated with MPTP (30 mg/kg, i.p.) daily for 5 days. QNC was injected i.p. in the doses of 0, 10, 30 and 60 mg/kg daily 30 min before MPTP in four different groups. Two other groups of mice received either vehicle (control) or a high dose of QNC (60 mg/kg). Two hours after the last injection of MPTP, striata were collected for the analysis of dopamine (DA), 3,4-dihydroxyphenylacetic acid (DOPAC), homovanillic acid (HVA) and glutathione (GSH). For the 6-OHDA study, male Wistar rats were infused with 6-OHDA (60 microg) in the right striatum under chloral hydrate anesthesia. The rats in different groups were treated with 0, 5, 15 and 30 mg/kg QNC (i.p.) for 4 days, while first injection was given 30 min before 6-OHDA. On day 5, rats were sacrificed and striata were stored at -80 degrees C. Administration of MPTP or 6-OHDA significantly reduced striatal DA, which was significantly attenuated by QNC. Concomitant treatment with QNC also protected animals against MPTP or 6-OHDA-induced depletion of striatal GSH. Our findings clearly suggest the role of PLA2 in MPTP and 6-OHDA induced neurotoxicity and oxidative stress. However, further studies are warranted to explore the therapeutic potential of PLA2 inhibitors for the treatment of Parkinson's disease.

B Vitamins Supplement Potentiates Antiparkinsonian Effect of Flunarizine: the Behavioral and Biochemical Evidences From 6-Hydroxydopamine Animal Model

Introduction: Prominent data indicate that flunarizine (flu), a calcium channel blocker, has neuroprotective effect. However, several authors have reported that the chronic use of flu can produce drug-induced Parkinsonism. Previously, we showed that B vitamins supplement (B com) has antiparkinsonian effect. In the present study, we evaluated the effect of pretreatment with flu ...

Sleep deprivation disrupts striatal anti-apoptotic responses in 6-hydroxy dopamine-lesioned parkinsonian rats

Objective(s): The present study was conducted to examine the effect of sleep deprivation (SD) on the anti-apoptotic pathways in Parkinsonian rats. Materials and Methods: Male Wistar rats (n = 40) were assigned to four groups (10 animals each): sham surgery (Sham), 6-hydroxydopamine (6-OHDA)-lesioned (OH), 6-OHDA-lesioned plus grid control (OH+GC), 6-OHDA-lesioned plus SD (OH+SD). Determination of the Role of Synapse Associated Protein 97 (SAP97) in the Normal and Parkinsonian Striatum By

Parkinson’s disease (PD) is a debilitating movement disorder associated with the death of dopaminergic nigrostriatal neurons. In addition to dopamine deficiency, abnormalities in glutamate and other receptors at striatal synapses have been reported. Synapse associated protein 97 (SAP97) is involved in regulation of glutamate receptor function. In the striatum of unilaterally-lesioned 6-OHDA rat...

اثر دویدن روی نوارگردان بر سطح IGF-1 در جسم مخطط موش‌های پارکینسونی شده با القای 6-هیدروکسی دوپامین

Background & Aims: There is no agreement on the protective effect of exercise on neurodegeneration processes, and the mechanisms responsible for exercise benefits in Parkinson disease (PD) yet. In this study we investigated the protective effect of 4 weeks of treadmill running with low intensity on the IGF-1, dopamine and tyrosine hydroxylase levels in the striatum of parkinsonian rats ind...

The Protective Effect of Voluntary Exercise on the Hippocampal Cerebral Dopamine Neurotrophic Factor Level against Intraventricular Injection of 6-hydroxydopamine in Rats

Background & Aims: The purpose of this research was to study the protective effect of pretreatment with a voluntary exercise on hippocampal level of cerebral dopamine neurotrophic factor (CDNF) after damage induced by intraventricular injection of 6–hydroxydopamine (6-OHDA) in rats. Methods: In this experimental study, 24 Wistar rats were randomly divided into 4 groups of healthy control, healt...
